A practical culture technique for enhanced production of manganese peroxidase by Anthracophyllum discolor Sp4 BABT

In this study, different growth conditions of Anthracophyllum discolor Sp4 including the effect of agitation, additions of lignocellulosic support, inducer and surfactant were evaluated on the MnP production in Kirk medium using a culture system made up of the tubes containing the glass bead . The highest MnP production (1,354 U/L on day 13) was obtained when the medium was supplemented with wheat grain and 0.25 mM MnSO4 as inducer, under static conditions at 30°C. Two isoenzymes were purified (35 and 38 kDa respectively). MnP presented a maximal activity in the pH range between 4.5 and 5.5, a relatively high temperature tolerance (50ºC) and a high catalytic activity for 2,6-dimethoxyphenol and hydrogen peroxide.

Worldwide use of pesticide has increased dramatically during the last two decades. As a consequence, pesticide residues and their transformation products are frequently found in groundwater and surface waters. This review summarizes information about polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs), some chlorophenols; it mainly empathizes on pesticides, their incorporation into the environment, microorganisms involved in their degradation and, some physico-chemical aspects of pesticides behavior in soils. This research aimed to evaluate the potential of several native white rot fungi (WRF) isolated from subtropical environments of Misiones (Argentina) to produce different ligninolytic enzymes. Coriolus versicolor f. antarcticus BAFC 266, Pycnoporus sanguineus BAFC 2126 and Phlebia brevispora BAFC 633 showed the highest phenoloxidase activity. Ganoderma applanatum strain E, P. sanguineus BAFC 2126 and P. brevispora BAFC 633 revealed marked laccase and peroxidase activity. ABSTRACT Lentinus crinitus is a white-rot fungus that produces laccase, an enzyme used for dye decolorization. Enzyme production depends on cultivation conditions, mainly agro-industrial by-products. We aimed to produce laccase from Lentinus crinitus with agro-industrial by-products for dye decolorization. Culture medium had coffee husk (CH) or citric pulp pellet (CP) and different nitrogen sources (urea, yeast extract, ammonium sulfate and sodium nitrate) at concentrations of 0, 0.7, 1.4, 2.8, 5.6 and 11.2 g/L. Enzymatic extract was used in the decolorization of remazol brilliant blue R. CH medium promoted greater laccase production than CP in all evaluated conditions. Estudio de la actividad lignocelulósica del hongo Pleurotus ostreatus sobre el rastrojo de cebada Colegio de Postgraduados

Este trabajo describe la evaluación del crecimiento del hongo ligninolítico Pleurotus ostreatus (cepas IE8 y CP50) sobre rastrojo de cebada suplementada con fuente de nitrógeno exógena (urea o sulfato de amonio) y humedades de 70 y 80 %; inicialmente se evaluó la velocidad de crecimiento radial (Vr) en cajas Petri y rastrojo de cebada. Las Vr más altas fueron 0.568 mm h-1 del testigo A (CP50, 80% de humedad, Medio con agua destilada) y 0.573 mm h-1 del tratamiento 6 (cepa IE8, 80% de humedad, Medio base). (Ver cuadro 5, pag. 23). Pellets of the white-rot fungus Anthracophyllum discolor with activated carbon and sawdust (complex pellets) were formulated for the degradation of lignin in water and the bioremediation of soil contaminated with pentachlorophenol (PCP). The complex pellets were formed by a center of activated carbon and sawdust surrounded by fungal mycelium, whereas simple pellets consisting of only mycelium were hollow spheres. Degradation of lignin was performed in an airlift reactor at initial lignin concentration of 1000 mg L-1 and the fungal pellets were re-used two times in batch operations. Production and characterization of tyrosinase activity in Pycnoporus sanguineus CCT-4518 crude extract BJM

Tyrosinase is an enzyme of industrial interest. The production and characterization of tyrosinase from P. sanguineus CCT-4518 were investigated. The selection of inductors, luminosity influence, inoculum size and type of culture medium on the production of tyrosinase and the effect of inhibitors on enzyme activity were performed. Optimum conditions for intracellular tyrosinase production was observed after 2 days using 0.15% L-tyrosine as inducer, in the presence of light, with inoculum size of 10 mycelium discs, using 2% malt extract broth medium, incubated at 30°C, and constant agitation of 150 rpm. Six strains of white-rot fungi isolated from southern Chile were evaluated for their ergosterol/biomass correlation and ligninolytic potential in solid medium to formulate pellets for Reactive Orange 165 (RO165) decolourization. The fungus Anthracophyllum discolor was selected to formulate complex pellets (fungal mycelium, sawdust, and activated carbon), coated pellets (complex pellet + alginate) and simple pellets (fungal mycelium).
